Removing Custom Columns/Stats/Variables

What's the best way to remove all custom columns/stats/variables so that only the defaults are available?

Move the C:\Program Files\PokerTracker 3\Data\StatsDefinitionsCustomized.pt3 file to somewhere else and restart PT3. That file contains all of your custom stats and if you move it to somewhere else and restart PT3, PT3 won't know about it and thus you'll start out clean.

Note, though, that if you've used any custom stats in your HUD layout, they won't be removed from your layout in this fashion. You'd want to also move the Layouts.pt3 file to get back to a default layout.
